چکیده

Glutamate synthase (GltS, EC 1.4.1.13) forms with glutamine synthetase (GS, EC 6.3.1.2, reaction 1) the main route for glutamate synthesis in micro-organisms by catalysing the NAD(P)Hdependent transfer of the amide group of glutamine to the C(2) carbon of 2-oxoglutarate (2-OG, reaction 2). The overall reaction catalysed by GS and GltS (reaction 3) is similar to the reaction of glutamate dehydrogenase (reaction 4), except for the expenditure of an extra molecule of ATP for each glutamate that is produced. Amidotransferases then can make this nitrogen atom available for amino acid biosynthesis.

Adenylylsulphate reductase from the sulphate-reducing archaeon Archaeoglobus fulgidus: cloning and characterization of the genes and comparison of the enzyme with other iron-sulphur flavoproteins.

Adenylylsulphate (adenosine-5'-phosphosulphate, APS) reductase from the extremely thermophilic sulphate-reducing archaeon Archaeoglobus fulgidus is an iron-sulphur flavoprotein containing one non-covalently bound flavin group, eight non-haem iron and six labile sulphide atoms per molecule. Reevaluation of the enzyme structure revealed the presence of two different subunits with molecular masses...

Purification and characterization of glutamate synthase from Azospirillum brasilense.

Growth conditions for Azospirillum brasilense Sp6 were devised for maximal expression of glutamate synthase. The enzyme levels were largely affected by the type and concentration of the nitrogen source. A 10-fold increase in the synthesis of the enzyme was observed at a limiting concentration of ammonia. The enzyme was purified to homogeneity by a procedure which was fairly rapid and allowed a ...

Purification and properties of glutamate synthase from Nocardia mediterranei.

Glutamate synthase was purified about 250-fold from Nocardia mediterranei U32 and characterized. The native enzyme has a molecular weight of 195,000 +/- 5,000 and is composed of two nonidentical subunits with molecular weights of 145,000 +/- 5,000 and 55,000 +/- 3,000. This enzyme is a complex of iron-sulfur flavoproteins with absorption maxima at 278, 375, 410, and 440 nm. It contains 1.1 mol ...

iron-sulphur protein subunit of the succinate dehydrogenase of Escherichia coli

The nucleotide sequence of a 961 base-pair segment of DNA containing the sdhB gene, which encodes the iron-sulphur protein subunit of the E. coli succinate dehydrogenase, has been determined. The sdhB structural gene comprises 711 base pairs (237 codons, excluding the translational initiator and terminator). It is separated by a 15 base-pair intergenic region from the preceding flavoprotein gen...

Molecular recognition in protein complexes involved in electron transfer.

Introduction Electron transfer reactions involving protein complexes are important in many biological processes, including photosynthesis, respiration, nitrogen fixation and steroid synthesis. The interprotein electron transfer reaction occurs either by orbital overlap or through a number of chemical bonds in the polypeptide chain that act as pathways for the electrons to communicate with the t...
